Just wanted to share the Lyme-disease controversy :


Up to one third of Lyme disease patients who have completed a course of antibiotic treatment continue to have symptoms, often termed "post-Lyme syndrome", such as severe fatigue, sleep disturbance, unconsciousness, and cognitive difficulties, with these symptoms being severe in about 2% of cases.[8][9] While it is undisputed these patients can have severe symptoms, the cause and appropriate treatment is controversial. The symptoms may represent "for all intents and purposes" fibromyalgia or chronic fatigue syndrome.[10] A few doctors attribute these symptoms to persistent infection with Borrelia, or coinfections with other tick-borne pathogens, such as Ehrlichia and Babesia.[11][12] Other doctors believe that the initial infection may cause an autoimmune reaction that continues to cause serious symptoms even after the bacteria have been eliminated by antibiotics.[13]

So apart from Chronic Fatigue, Post-Finasteride Syndrome, Post-Accutane Symptoms we add a new Syndrome called Post-Lyme Disease Syndrome (PTLDS). According to my Theory All conditions have the same basis, namely Oxidative/ER Stress dysregulation which originates from Liver Injury. This coupled with several SNPs that limit ? (hypothesis) the Redox Potential of an individual leads to a vicious cycle of Oxidative - ER Stress.

Abstract
Hypothalamic-pituitary-adrenal (HPA) axis dysfunction has been found in a high proportion of chronic fatigue syndrome (CFS) patients and includes enhanced corticosteroid-induced negative feedback, basal hypocortisolism, attenuated diurnal variation, and a reduced responsivity to challenge. A putative causal role for genetic profile, childhood trauma, and oxidative stress has been considered. In addition, the impact of gender is demonstrated by the increased frequency of HPA axis dysregulation in females. Despite the temporal relationship, it is not yet established whether the endocrine dysregulation is causal, consequent, or an epiphenomenon of the disorder. Nonetheless, given the interindividual variation in the effectiveness of existing biological and psychological treatments, the need for novel treatment strategies such as those which target the HPA axis is clear.


Another potential cause of disruption to HPA axis function is oxidative stress and a decrease in antioxidant capacity in addition to the presence of histone deacetylase (HDAC) [72]. Increased activity of HDAC 2 and 3 coincides with a decrease in plasma cortisol [14]. This theory has been cited as another possible cause of hypocortisolism found in patients.

just a small update.So re-dosed the choline/inositol combo product, as opposed to just the choline about an hour ago. Just a calm subtle mood boosting effect, and none of the sides I experienced with just choline alone. Idk what the deal with that is but maybe im low on inositol? Lol is that even possible? I guess theres nothing normal in this disease, though, so Im kind of used to weird reactions to things.

To be honest i do not know. There are some enzymes that use Inositol and have functions for ER Stress signalling such as IRE1 (inositol-Requiring Enzyme 1).

I am glad you are feeling better.

Some comments :


-Your subclinical Hypothyroidism originates from HPA Dysregulation due to Oxidative Stress (my Hypothesis)

-Your elevated bilirubin levels *may* indicate liver damage. Consider taking TUDCA.If you cannot take TUDCA at present, try taking Taurine 500 mg every day and evaluate. Since Liver function is important (My Hypothesis) try adding this first (ie TUDCA or Taurine)

-Avoid P450 Inhibitors, as discussed at the beginning of this Thread
-Consider adding P5P to your regimen and evaluate how you feel.
-Try also adding Molybdenum (small amount, ie 50 mcg ) to your regimen and evaluate
-For every supplement you are adding, evaluate for a week THEN add a second (if needed)

It is not clear how much Choline/Inositol you are taking, could you let us know?

After a month i suggest you re-test bilirubin and your Thyroid function. The test that i have done to evaluate my Thyroid function is called 'TRH'. If you find your TSH to be considerably lower then this will be a good sign.

This is interesting, some viruses actually inhibit hsp 70! Quite the opposite of what EBV does.

This paper will address consequences to hosts when the Hsp stress response is impaired due to viral infection. (My add in: PEM!!!)
It was also recently demonstrated that HSV-1 inhibits the unfolded protein response (UPR) during the early stages of infection of cultured human cells. Viral inhibition of UPR frees the virus from fidelity control of protein synthesis and can increase the efficiency of viral replication 1,000-fold (Burnett et al. 2012; Lutsenko et al. 2010). Loss of UPR disrupts endoplasmic reticulum homeostasis and can thereby lead to a number of pathological sequelae (see below).

I Just had an interesting "flu" which lasted only one day. Weather condition where i am are so bad, we even had a Twister in the city.

Trial Update Day 60

In the following, changes from my last update will be in bold.

I'm currently taking,
  • 3x1400 mg NAG
  • 0 mg TUDCA: discontiniued on day 51
  • 0 mg FMN ( B2 ) : discontinued : I had tried FMN before and found no benefit over the B2 I get in a b-complex. The same appears to be true now.
  • 100 mcg selenium
  • 3x250 mg citicoline:
  • 3x300 mg alpha GPC
  • 5x500 mg choline (as choline L-bitartrate) : bumped up from 3x500 mg
  • 3x750 mg inositol : bumped up from 750 mg
  • 3x100 mg Pantothenic Acid ( B5 )
  • 2x300 mg Pantethine : new addition
along with methylation and mineral supplements. I have also been on the autoimmune paleo diet for 44 days.

Improvements:
  • 50% lightening of sun damge/ liver spots on forearm ( onset one year ago )
  • decreased urinary frequency. Now I go every 2-3 hours vs every hour
  • lighter urine color
  • better urinary flow ( probably due to less prostate inflammation )
  • 60% improvement in folloculitis on arms and legs ( onset one year ago ) :
  • 100% improvement in noise intolerance : up from 80%
  • 95% improvement in headaches.
  • 70% improvement in lower back pain ( onset one year ago ): down from 95%
  • 33% improvement in energy levels
  • 60% improvement in groin pull injury (onset 5 months ago, little improvement over previous 4 months )
  • 50% improvement in nasal congestion : up from 33%
  • POTS: Poor man's tilt table test results: 30-35 bpm :This is slightly down from the 30-40 bpm average that I posted last time, and a small improvement over the 35-40 bpm baseline when I started the trial.
No change:
  • eye floaters ( onset one year ago )
  • skin texture/ wrinkling ( onset one year ago )

Let's start with the bad news. After 4 days of using bulk powder choline bitartate, in the form of choline DL bitartrate, I began having increased lower back pains. I suspect the back pain might be related to this racemic form of bitartrate. I outlined my concerns in this post. The pain greatly subsided a day after I stopped using it and is nearly gone now. I've got a new bag of choline L-bitartrate that I've been using for a couple days. We'll see what happens.

My OI/POTS remains largely unaffected. The large doses of choline/B5/pantethine are an attempt to raise levels of acetylcholine in the hopes that this somehow corrects the OI problems. So far, no dice. But I'll continue the experiment for a while.

At this point, I don't think I'm any worse for discontinuing TUDCA. I'll be ready to order it again if I experience a crash, but so far so good.

Now the good news. Noise intolerance is gone! The Noise intolerance and headaches, when I have them, come along with a really nasty mood. I won't try to explain in detail except to note that both of these factors are really indicative of my overall mental well-being. When they are absent, I feel a hell of a lot better.

Although the noise intolerance has been slowly improving, due in large part to NAG I believe, what finally killed it was Pantethine. An hour after I took my first dose, it was gone. And it hasn't returned. We might theorize that Pantethine was the last ingredient I needed to bump up my acetylcholine levels, which in turn is responsible for fixing the noise intolerance.

( Keep in mind, it has only been 4 days since I started Pantethine. As we know these symptoms have a way of creeping back. I'll continue to update.)

BTW Have you ever had a complete Liver panel ?

If you haven't can you check ALT/AST, Albumin, Bilirubin,Gamma-glutamyl transpeptidase (GGT), Lactate dehydrogenase(LDH) and alkaline phosphatase (ALP) Levels ?

If any of these are elevated then you have issues with the liver which -in my opinion- should be addressed.

Have you completely stopped Alcohol Intake? If not, can you please try it for a month and evaluate how you feel?


Regarding acetylcholine and Coenzyme a, the matching is problematic because both "acetylcholine" and CoA exist as words in multiple versions so we have many partial matches which change the results.

If you haven't can you check ALT/AST, Albumin, Bilirubin,Gamma-glutamyl transpeptidase (GGT), Lactate dehydrogenase(LDH) and alkaline phosphatase (ALP) Levels ?

I don't have GGT or LDH. Everything else you list was within range when it was tested earlier this year. The only thing notable was that Bilirubin was at the very top of the acceptable range. The Doc told me I was perfectly healthy.

Have you completely stopped Alcohol Intake? If not, can you please try it for a month and evaluate how you feel?

Ha ha! Drinking and me don't mix anymore, unfortunately. I think I've had two beers all year and both were mistakes. I haven't had anything to drink during this trial.
