A Study to Evaluate ASP0367 in Participants With Reduced Maximum Oxygen Uptake Due to Poor Systemic Oxygen Extraction

Interesting new trial by Dr Systrom in collaboration with Mitobridge.

Why treatment of Poor Systemic Oxygen Extraction might be interesting for ME/CFS

References:
https://www.healthrising.org/blog/2...-exertional-malaise-chronic-fatigue-syndrome/ Systrom found that the muscles of 45% of the people in his study – all of whom met the criteria for chronic fatigue syndrome (ME/CFS) – were unable to extract normal amounts of oxygen from their blood. The inability to extract enough oxygen from their red blood cells could explain why people with ME/CFS have so much trouble with exercise; i.e. their muscle cells simply aren’t getting the fuel (the oxygen) they need to engage in exercise. The oxygen extraction issues were found even during rest.

So if I’m understanding correctly, the purpose of the drug is to push beta-oxidation; how will this help if the underlying issue in ME is inadequate oxygen delivery to cells?
PPAR detla seems to increase oxygen uptake in cells.
Systrom studies found that oxygen delivery/supply is not the problem, but impaired uptake capacity.
At least in 20-25% of his patients.

This would also match with our groups experience that ITPP increased oxygen supply does not help severe-very severe patients but only mild - moderate (those seem to have still working oxygen uptake and might thus benefit from increased oxygen supply of ITPP)

However I would love if the study site would provide any info on what results they expect and why. Couldnt find any official data tho.

Thank you #mitoMAN for posting this. However, when I see company-sponsored research showing great results, I don't just take it with a grain of salt -- I grab the whole salt shaker.

only way to find out is either take it ourselves or wait until 2023-2025 when study data from the clinical trial is published

Thanks for posting this as I find it very interesting knowing that I get benefit from breathing oxygen from my concentrator, at least 3 times daily for around 25 minutes. I can feel it putting back energy into my muscles especially first thing in the morning, after lunch or after any physical activity that leaves me feeling worn out.

I have been using a concentrator for 11 years now as it was prescribed by an ME clinic after a test on my autonomic system (just taking 6 breaths in a minute whilst they recorded the oxygen in my cells and my CO2 level). I was told my reading was very low and CO2 was a bit high and this was extremely bad for me, hence me getting the concentrator.

TBH I am not sure at all whether this relates to the study mentioned!

I will never forget the effect I noticed immediately after using the concentrator for the first time for an hour. When I took my dog for a short walk I couldn't get over how all the colours of the trees were so bright and sharply defined, it was like being in a different world. I am pleased to say that effect has stayed as I look around on my short walks but it goes to show what a lack of oxygen does to our brain, at least in my case.

@mitoMAN Could I get a discord invite? I've just purchased ITPP.
DMed you

Another interesting study finding supporting the use of PPAR Delta agonists to correct dysfunctional fatty acid oxidation.

https://www.healthrising.org/blog/2022/05/18/long-covid-exercise-cpet-mitochondria/

This study, “Decreased Fatty Acid Oxidation and Altered Lactate Production during Exercise in Patients with Post-acute COVID-19 Syndrome“, brings in a cadre of researchers from the Pulmonary Physiology Lab at the University of Colorado Anschutz medical campus, and, get this, the Center for Post COVID Care at National Jewish Health Hospital, in Denver. National Jewish Health, by the way, calls itself “the leading respiratory hospital in the nation and the only one devoted fully to the treatment of respiratory and related illnesses”.

@mitoMAN
I'd also love a Discord invite. This whole PPAR-delta angle seems really interesting. I'm also in the market for some ITPP, both for my CFS and for my girlfriend's beta thalassemia minor.

What are you thoughts regarding self decodes article warning about taking cardarine? https://drugs.selfdecode.com/blog/gw501516-cardarine/
You will find all the info in our discord.
Overall cardarine is only considered safe in human studies up to 4 months.
We are only trying cardarine for 1-2 months to evaluate if PPAR Delta is of interest for us.
If you want to take it long term, you need to synthesize Bocidelpar

Cool, I've signed up for the discord. See you over there soon hopefully.
