i always was wondering why this one certain diabetes drug made me feel so much better, its name is pioglitazone also known as actos.
i am SOD2 homozygote.

i did AI work today, and i found this:
https://selfhacked.com/blog/the-brain-fog-gene-rs4880-or-sod2/

PPAR Gamma is helping in SOD2 , so pioglitazone increases PPAR Gamma. its one of its main targets and functions.

so could be pioglitazone a potential treatment for SOD2 homozygotes?

I'm not sure what my SOD2 type is but apparently cinnamon also does this which helps me a lot, unlike anything else and I couldn't explain it after digging around so much but I'm wondering is this may be it. Stacking high dose vitamin C on top of it (I take 2 grams every morning now) has been a game changer for me which also scavenges SO. With this on my current supplement stack I hang around mild most of the time with some neuro-cognitive issues.

Few questions . Do you have CFS according to fukuda criteria I mean do you have pem. 2nd what does if pioglitazone you used.
3rd on a scale of 1 to 10 how much you improved?

i dont have cfs, i do have pem, 15-30mg , should have started with 15. doctors were overeager.
i cant do the scale, i only know that my exercise tolerance increased a lot, lets say instead of a bad day i have a good day. my glucose levels improved as did my liver values. but it only worked for a few month. then it was normal, i got worse when i didnt take it. i slowly got down from 30 to 15.. then 22,5 ... when i have a hard day i use 30mg as pem blocker

PEM only occurs in ME/CFS Or fibro. if you have pem then it's CFS/me Or fibro

i dont believe in CFS being a disease, i believe its a symptom group with many different diseases causing it, in my case a metabolic myopathy or mitochondrial disease might be the origin.
but a homozygote SOD2 plus a few other factors could also lead to mitochondrial dysfunction. sometimes you got 1 or 2 bad genes and its fine, but then comes another 3rd or environmental factors and the body is overwhelmed and declines.
despite many others with cfs, i did not get it out of nothing, i have problems since i was a kid. and it slowly got worse and worse.

How, you found your root cause?

How, you found your root cause?
not exactly, thats what doctors suspected, but they couldnt find exact disease. and they do not want to continue, tell me i shall do expensive gene testing, but insurance doesnt pay much and doctors actually do not know for what to look.
i did a private whole genome sequencing, and didnt find a exact cause, but a expert would need to analyse it.

Are you aware of Hips thread about using B12 oils? The guy who makes those who is a biochemist I believe will analyse your data for free, last time I checked, he is typically just interested in peoples stories and maybe data as well. There is also a guy called Dr. Dan Purser who I follow on YT, he seems to have quite an extensive knowledge on these subjects and puts out a lot of content, on specific snps, supplements and such, he mentions SOD quite a bit. I am just slightly sceptical with the supplements he has and seems to push quite a bit, I am not sure you treat SOD with more SOD neither. But he might actually be worth considering a tele appointment with, or his staff, since overall they seem very genuine.

I am concerned about my SOD1 these days, as I seem to have more connective, muscular and signalling issues than ever, but haven't been pursuing what to do about it really neither.

God, I just found the prices for appointments with Dr. Dan, unreal 😏

I am trialing pioglitazone 15 mg. Bought 30 tablets. Here pharmacist give any medicine to patients.
hope it goes good, remember its a diabetes medicine pushing down insulin resistence with potential side effects like gallbladder issues, weight gain, heart weakness etc.
i would not go above 15mg and if possible even go down to 7,5.
