Il-2 Receptors/CD25

Has anyone had Interleukin 2 Receptors tested? Mine was done 2 years ago and came back: <406 with range 406-1100. The neurologist who ordered said this indicated that my immune system was really busy fighting or something like that, but earlier this year a naturopath said that was an immune deficiency.

I forgot all about it until I came across the test result page a few moments ago and googled it. Wikipedia says "CD25 deficiency or interleukin 2 receptor alpha deficiency is an immunodeficiency disorder associated with mutations in the interleukin 2 receptor alpha (CD25) (IL2RA) gene. The mutations cause expression of a defective α chain or complete absence thereof, an essential part of high-affinity interleukin-2 (IL-2) receptors. The result is a syndrome described as IPEX-like[1] or a SCID.[2]"

Although I have had some odd sort of things pop-up, I don't think I have anything IPEX-like or SCID. So, I ask: Have you had Il-2 Receptors tested? Is this common in PWME?

Hi Isaiah, did you check your IL2RA gene?

I went on the wikipedia page for the SCID syndrome you are talking about but couldn't find anything about this IL2RA gene...

I searched also for IPEX syndrome, and found a mutation in the FOXP3 gene, leading to lymphocyte T reg dysfunction

edit: I checked my genes and found a missense mutation on the PNP gene (considered as benign)

and for the IL2RA gene, variants have shown increase risk for some auto-immune diseases:

Rheumatic arthritis, Juvenile idiopathic arthritis, Multiple Sclerosis, Graves disease, type I diabete...

in this study about haplotype at the IL2RA gene and risk for Diabete:

"We demonstrated that the presence of an autoimmune disease-associated IL2RA haplotype correlates with diminished IL-2 responsiveness in Ag-experienced CD4(+) T cells, as measured by phosphorylation of STAT5a, and is associated with lower levels of FOXP3 expression by Tregs and a reduction in their ability to suppress proliferation of autologous effector T cells. These data offer a rationale that contributes to the molecular and cellular mechanisms through which polymorphisms in the IL-2RA gene affect immune regulation, and consequently upon susceptibility to autoimmune and inflammatory diseases."

So an IL2 receptor deficiency can be associated with increase risk for some auto-immune diseases. I don't know if this parameter where studyed among the CFS/ME patients, it would be interesting!

@Isaiah 58:11 , do you know your snp for the IL2RA gene?

I have the IL2b Low apparently very common low on CFS (according to my doctor), In all my treatment has not gown up for me. I do not know anything about CD25. Since usually they CFS doctors associate CFS with CD56 or CD57(lyme) CD3 (I think Sojern). They did test me for it but was not like a big deal. So cannot help you with that one.

I think in my case, they check the overall lymphocyte to see how my immune system is doing (over, under, unregulated...) they have me on immune modulators (Working for me, not sure but has me stable).

That is good to know, thank you. I will see an ME/CFS specialist for the first time next month and hope to get caught up on what is "normal."

Interesting. I need to re-read that book to take notes when able. It was a rather traumatic experience for me teh first time through. I came away thinking that I would end up a wig-wearing dementia patient in a wheelchair, which was/has been awfully close to my experience.

I had that test done 11 or 12 years after onset. I wonder if it used to be high and then fell. I think I recall that some markers have shown to do that?

Mine are both low… IL-2 and CD4+CD25+ and lastly, the cytotoxic CD8+T cells.

My CD4+CD25+ count was literally 1/mm3 out of the standard 52-255 range.
