LPS impairs the brain's ability to get rid of waste

Lipopolysaccharide impairs amyloid β efflux from brain:
altered vascular sequestration, cerebrospinal fluid reabsorption, peripheral clearance and transporter function at the blood-brain barrier.

...Our findings that systemic inflammation causes deficits in both Aβ transport and bulk flow like those observed in AD indicate that inflammation could induce and promote the disease.

Hi ebethc- Dr. David Perlmutter has researched this extensively and talks about the connection of dysbiosis and leaky gut to AD often. His father passed away from AD, so he is very much on top of the research.

How do you know if you're making any progress healing leaky gut? It goes so slow, and there are so many problems it causes (which also have to be fixed) that's it's hard to know... For example, is there a test for LPS levels so you can track levels dropping over time? wouldn't that be nice

I think the lactulose/mannitol test is the gold standard. I've never had one, though.

EDIT: my opinion on this is that it is the microbiota that matters most, i.e. when the microbiota is normalized leaky gut will take care of itself. So I would focus on the microbiota, not leaky gut per se.

so, how do you know that your microbiota is improving? is there a test for that?

EDIT: SIBO just tests whether bacteria is in the wrong place, not whether you have the right amount of the good stuff, or reduction in bad.... uBiome?? it doesn't seem that accurate

There was also this study published around the same time which suggested that liver LRP played a key role in clearing amyloid in the brain.

Withania somnifera reverses Alzheimer's disease pathology by enhancing low-density lipoprotein receptor-related protein in liver
A 30-d course of oral administration of a semipurified extract of the root of Withania somnifera consisting predominantly of withanolides and withanosides reversed behavioral deficits, plaque pathology, accumulation of β-amyloid peptides (Aβ) and oligomers in the brains of middle-aged and old APP/PS1 Alzheimer's disease transgenic mice. It was similarly effective in reversing behavioral deficits and plaque load in APPSwInd mice (line J20).

Withania somnifera is commonly known as ashwagandha.

I struggle with knowing how much I have improved my gut too sometimes. I try to remember all the things that have improved and sometimes it's not easy! I clearly have come a long way in 4-5 months, however, that is very easy to loose sight of.o_O

I feel like the other problems that it causes, at least what I believe it causes, like mito dysfunction, immune system dysfunction and possibly neuro-inflammation, are almost impossible to figure out, for me anyway. They are often very distracting but on the other hand, I think treating them too could be very helpful, especially immune system imbalance. It does get very confusing sometimes!!

I am certain immune system dysfunction causes me mild to moderate flu-like symptoms. This morning was a good example. Shortly after I woke up, I felt like I was catching a flu but it wasn't a flu. So I went up on the dose of cordyceps I usually take and within an hour, the flu-like symptoms were almost gone! I have done this many, many times.

However, if I take too much, I will feel worse, which seems to suggest that I have over-shot my immune system balance. So it seems that an under-active immune system or an over active immune system both cause the same flu-like symptoms.

I have read many papers that talk about a healthy immune system needing to be able to go up and down to address pathogens etc, as is needed to maintain balance. My immune system don't seem to be able to maintain that balance, which causes my flu-like symptoms. Anyway, that's my story and I'm sticking to it, for now.:D

I think there are tests for LPS levels in the blood but I don't know if just any doctor can order them or if it needs to be a specialist.

what's LRP?

I started taking Wobenzym last Fall and after a couple of days I got a massive sinus headache, then it subsided and it's only been helpful since then.... I wonder if the enzymes broke up plaque, and that's what caused the headache?? I think that's what happened

I can't take anything that affects thyroid.... maca, ashwagandha, etc.

I'm waiting for the results of my first uBiome test now. No tests are perfect, but at least uBiome can show the level of diversity and ratios of different species. I think it's the best we have for now.

interesting... keep us posted

how do you detox the brain (beyond breaking up the plaque, which is great..)? everytime the pollution goes up slightly I feel like I have cotton in my brain! I have a dull to severe pain in my sinuses, and my IQ drops 40 points... I've been taking high dose Vitamin C and that seems to help a little; mostly it helps recovery once the pollution has subsided, like late at night

EDIT: also, DHA and astaxanthin seems to help, but I think it's more likely that they're just bringing inflammation down (good) but not helping the glymphatic system remove any toxins

I've had the same experience.... a "wobbly" immune system! plus, I get every cold/virus that goes around, so in addition to the baseline flu symptoms, I get actual cold/viruses then have a hard time getting over them... It's really hard to sort out progress w these conditions.... I felt like I was getting better in May, then got a severe throat infection which I'm just recovering from... Not sure if the mushrooms (cordyceps, reishi, chaga) stimulated EBV (it sure felt like it) and therefore symptoms were caused by an existing virus surfacing for a last gasp before being driven into dormancy OR it was an altogether new illness... I tend to think the latter, but maybe that's just hopefulness on my part... The lack of metrics to track progress (or lack thereof) is exasperating.

I agree!! It's very frustrating. I feel like I'm doing great some days and I am doing very well, but then I will have a period of fatigue and think "I'm stuck and am not making any progress at all!"

Having tests like you mentioned above would be great! That way even if we didn't feel the progress, we could see it on paper.:)The really weird thing is now I know I can get out of those low energy periods, most of the time, just by increasing the immune modulators I take or the antibiotic herbs.

The biggest problem with that is I can easily become over-stimulated and not be able to sleep well, leaving me fatigued the next day. It's like trying to drive a truck through the eye of a needle!:D

what's LRP?

LRP stands for low density lipoprotein receptor-related protein and it is involved in transport and degradation of various substances. Both the LPS study that you linked and the ashwagandha study that I linked suggested that it is involved with amyloid deposit and clearance.

What I find interesting in the ashwagandha study is that it suggests that liver LRP plays the key role in amyloid clearance. So to remove amyloid from the brain, you don't need substances to cross the blood-brain barrier, you just need to work on the liver. The same would conceivably hold for other toxins.

interesting.... what has worked for you to remove toxins from liver? there's an endless amount written, and I've tried many, but still looking for tips.... The usual suspects (milk thistle, turmeric) do nothing; lemon juice, bupleurum & burdock are good for me.

ashwaganda is out due to impact on thyroid... maybe a super low dose?

You can test soluble CD14, that seems to be a test voor LPS in the body.
Zonuline is a test for leaky gut.
