Thanks, GG - I've got OI, not sure if it's low-flow POTS. This is an interesting article. Does anyone know what the IV dose of Vit C in this study would correspond to as a dose of oral Vit C?

I didn't understand what the IV dose was? Help? I have taken a lot of IV vit C infusions and never noticed its affect on OI. I don't have POTS though, but NMH.

You didn't understand because they didn't tell. 10 mM (millimolar) is a measure for solution concentration and not quantity. And they didn't take it intravenous but intradermal (if that matters).

Thanks heaps for posting this study here, Ive copied it and will take it to the specialist with me when I go and see one for the POTS next month.

I too dont know if I have "low flow" POTS or not thou I assume I may thou I dont get many of the signs I have low blood flow anymore so its all rather confusing.

I wonder how much longer vitamin C stays in the body with this method of delivery compared to taking it via mouth. (I know when taking it by mouth one basically pees it right out so hence you need more then one dose per day, several doses are far preferable).

"A 25 gauge needle is inserted into the dermal space and exits the skin approximately 15 mm distant. The microdialysis catheter is inserted into the lumen of the needle, and withdrawn so that the dialysis membrane is 10 mm from the point of insertion. Once the membrane is placed, the needle is withdrawn. Multiple catheters are typically used but must be spaced at least 6 cm apart throughout all procedures and are taped in place. "

(that microdialysis page also does have links to POTS stuff so I assume this gives us a good idea of what the deliverly procedure).

"To date, microdialysis is the only sampling technique that can continuously monitor drug or metabolite concentrations in the extracellular fluid of virtually any tissue" (cyte from wiki - http://en.wikipedia.org/wiki/Microdialysis).

So they used this method because it's the only way to administer and monitor vitamin C concentration simultaneously, and not because it's effective or something. Intravenous and intramuscular are much more effective, as I remember 70 times more vitamin C comes to blood compared to oral administration.

Abstract*

Low flow postural tachycardia syndrome (LFP) is associated with vasoconstriction, reduced cardiac output, increased plasma angiotensin II, reduced bioavailable nitric oxide (NO), and oxidative stress.

We tested whether ascorbate would improve cutaneous NO and reduce vasoconstriction when delivered systemically.

We used local cutaneous heating to 42C and laser Doppler flowmetry to assess NO-dependent conductance (%CVC(max)) to sodium ascorbate and the systemic hemodynamic response to ascorbic acid in 11 LFP patients and in 8 control subjects (aged 23 2 yr).

We perfused intradermal microdialysis catheters with sodium ascorbate (10 mM) or Ringer solution. Predrug heat response was reduced in LFP, particularly the NO-dependent plateau phase (56 6 vs. 88 7%CVC(max)).

Ascorbate increased baseline skin flow in LFP and control subjects and increased the LFP plateau response (82 6 vs. 92 6 control).

Systemic infusion experiments used Finometer and ModelFlow to estimate relative cardiac index (CI) and forearm and calf venous occlusion plethysmography to estimate blood flows, peripheral arterial and venous resistances, and capacitance before and after infusing ascorbic acid.

CI increased 40% after ascorbate as did peripheral flows.

Peripheral resistances were increased (nearly double control) and decreased by nearly 50% after ascorbate.

Calf capacitance and venous resistance were decreased compared with control but normalized with ascorbate.

These data provide experimental support for the concept that oxidative stress and reduced NO possibly contribute to vasoconstriction and venoconstriction of LFP.

Thank you Dolphin, any study into alleviating the awful and debilitating symptoms of pots gets my vote as I suffer terribly.

But :Sign Help:, can anyone interpret this study in plain speaking please? What did they do and how? And what might it mean for us potsy people in terms of taking Vit C?
