Hello - I recently discovered this site and am excited to find such a helpful and knowledgeable community. I've been researching natural ways to treat lifelong symptoms of depression, mood swings, anxiety and brain fog and am delving deeper into supplementation, methylation and genetic testing.

I received my results back from 23andMe; this is a complicated journey but I am trying to figure out the initial steps and treatments for these genetic anomalies. This forum (and MTHFR.net) have been excellent resources so far, but I am at a bit of a loss of what to address first! And as to what combinations of gene defects can be tricky. Any insights or suggestions for what to look into would be greatly appreciated.

My results are below; the ones that seem most concerning are MTHFR a1298c homo, MAO A R297R hetero, CBS 699T homo.

COMT V158M rs4680 GG -/-
COMT H62H rs4633 CC -/-

Symptoms: lifelong depression, anxiety and mood swings (worsened significantly for 1-2 weeks around my period/PMS-induced and debilitating at times). Brain fog, fatigue, low stamina; not all of the time, but more often than not and seem to correlate to my period, lack of sleep, stress, and occasionally caffeine consumption (16oz cup every day). Have tested for low iron previously. Tested low-ish for vitamin D, which I know is related to VDR?). I am 28/F, 120lbs.

Current daily supplements:
am/day:
-fish oil (minami 850mg omega 3, 45mg omega 6, 35mg omega 9)
- vit b12 (~125-250mg jarrow methylb12, which I believe is the proper form for MTHFR)
- vit c (500mg+)
- vit d (upped to 2,000 iu)
- few times per week: Thorne Mediclear Plus powder- which seems to have a lot of helpful ingredients in general and the proper forms of b12 and folate as well as molybedum)
pm/evening:
- evening primose oil (500mg, sometimes 1,000mg around period)
- zinc (50 mg as amino acid chelate)
- probiotics (either udo's choice or pb8)
- magnesium/calcium blend (Solaray ~700 mg mag citrate/oxide and cal citrate/hydroxide).

I also take effexor 75mg daily and the occasional adderall xr 5mg (the latter makes me feel like a real person!). For diet, I focus on protein, fats, and am low-gluten, low-grain, low-sugar but have some slip ups. I eat greek yogurt every day and drink a lot of kombucha & mineral water.

Thank you in advance for any insights and I look forward to speaking to more of you on this forum!

Hey goonie. To me it looks like you're already all set regarding your mutations.
And I hope the stuff you take has a great positive impact.
As for your Brain fog, fatigue and low stamina you might want to look into supplements which increase your energy metabolism. In the brain as well as in the body.
Here are just a few keywords: Piracetam, Lipoic acid, Creatine

Good luck!

@goognie8473 - You look like you have my genes and I had all of your issues. Except the PMS cause I am a dude.

You need to be careful about the coffee, but not about the caffeine. Coffee has a HUGE amount of riboflavin. Roboflavin is great for MTHFR but it speeds up the Methionine cycle and effects the CBS SNP by producing too much homocysteine and therefore too much action down the transulfuration pathway. Trust me on this, you need to make sure you fix the CBS SNP first of you will be frustrated.

I would add pyridoxal 5 phosphate (B6) for that CBS SNP. Does wonders for me. I need to take B6 before I drink decaf coffee of I get brain fog and rhinitis badly. In fact it is the first subligual I take in the morning and my day is great.

I was hospitalized twice with severe anxiety and depression. It has been 10 years since I have been hospitalized and three months since I have had any depression or anxiety and I am on zero pharmaceutical meds. That is huge for me. I credit this this to understanding my SNPs.

As far as the addreal is concerned, yes, speed will always make most people feel better. ;)

Apologies for the late reply, but thank you both for the very helpful suggestions!

Methyl Heath, that is fantastic to hear that you have addressed your depression/anxiety and are no longer on the meddy-go-round. So many people struggle with those both severely and it's inspiring to hear that people can recover in ways that western medicine doesn't yet understand.

I will definitely try the p5p for CBS. And perhaps try switching to decaf coffee-- it is almost the same, right? :)
